iTRAQ (isobaric tags for relative and absolute quantitation) technology is an in vitro isobaric tagging technique for relative and absolute quantitation developed by AB SCIEX. It is a high-throughput screening technique commonly used in quantitative proteomics in recent years. iTRAQ utilizes multiple isotope reagents to label the N-terminal or lysine side-chain motifs of protein peptides, which can be analyzed in tandem with a high-precision mass spectrometer, allowing for simultaneous comparisons of protein expression between up to eight samples.
A sketch of the iTRAQ tag and the quantification method is shown in the figure below:
The technique uses four or eight isotopeencoded tags to specifically label the amino groups of peptides, followed by tandem mass spectrometry, allowing for simultaneous comparison of relative or absolute protein content in four or eight different samples. iTRAQ's four isotopelabeled reagents consist of four reporter groups with relative molecular masses of 114, 115, 116, and 117, a balance group with relative molecular masses of 31, 30, 29, and 28, and an identical peptidereactionlabeled reagent group. The four isotopic labeling reagents of iTRAQ consist of four reporter groups with relative molecular masses of 114, 115, 116, and 117, a balance group with relative molecular masses of 31, 30, 29, and 28, respectively, and an identical peptide reactive group. The relative molecular mass of the different reporter groups is 145 after matching with the corresponding balancing group, i.e., isobaric tag. The principle is that the protein is first cleaved into peptides and then differentially labeled with iTRAQ reagent.

1. Protein to be detected requirements: intact and not obviously degraded total protein, protein amount of a minimum of 300ug, the concentration of a minimum of 5ug/ul, using two-dimensional electrophoresis reagent extraction;
2.Provide tissues or cells directly, for cell samples, cell volume > 107; for tissue samples, wet weight of animal and microbial tissues > 10mg; for plant fresh tissues > 100mg; for body fluid samples, blood volume > 500ul; all the subsequent operations will be done by Tinker Biotech, and you only need to provide the samples, and no other reagents are required;
3.Protein transportation: can be transported at 4 degrees within 24 hours;
4.Protein long-term storage: -20 ℃.
