Differential fluorescence bidirectional gel electrophoresis (DIGE) is a method of labeling protein samples prior to 2D electrophoresis, allowing precise analysis of protein abundance between samples.
Ettan DIGE fluorescence differential protein expression analysis system combines the method of multiple fluorescence analysis based on the traditional bidirectional electrophoresis technique, which jointly separates multiple samples labeled by different fluorescence (Cy2, Cy3, Cy5) respectively on the same gel, and introduces the concept of internal standard for the first time, which greatly improves the accuracy, reliability and reproducibility of the results.
In DIGE technology, each protein spot has its own internal standard, and the software fully automatically calibrates the expression of each protein spot according to its internal standard, guaranteeing that the detected changes in protein abundance are real.DIGE technology can detect less than 10% differences in protein expression between samples, with a statistical confidence level of more than 95%. Ettan DIGE technology also allows proteomic analysis of micro-volume (as little as 5 μg) samples. The principle is as follows:
1, Protein to be detected requirements: intact and not obviously degraded total protein, protein amount of at least 5mg, concentration of at least 4ug/ul, extracted using two-dimensional electrophoresis reagents;
2、Provide tissues or cells directly, for cell samples, cell volume > 108; for tissue samples, plant fresh tissue, animal, microbial tissue wet weight > 300mg; for body fluid samples, blood volume > 1ml; all the follow-up operations will be done by Tinker Biologicals, and you only need to provide samples, and no other reagents are required;
3、Protein transportation: can be transported at 4 degrees within 24 hours;
4, Protein long-term storage: -20 ℃.
