Pseudoviruses are viruses in which a retrovirus is able to integrate the capsid glycoprotein of another virus to form a virus with an exogenous viral capsid, while the genome maintains the genomic properties of the retrovirus itself. Pseudoviruses can only infect cells once, have a wide host range, have high titers, and are less likely to be inactivated by serum complement than true viruses, and can be substituted for true viruses in neutralization assays. Some viruses, such as novel coronaviruses SARS-CoV-2 (2019-nCoV), SARS-CoV, Ebola virus and H5N1, are highly infectious and pathogenic, which bring great difficulties and dangers to studies such as screening for neutralizing antibodies. Therefore, it would be a very safe and effective research tool to screen antibodies or sera with neutralizing activity against pseudoviruses by packaging pseudoviruses, simulating the biological process of antibody blocking virus entry into cells, and detecting whether antibodies or sera have neutralizing activity against pseudoviruses.

At present, we have successfully packaged the new coronavirus pseudovirus with HIV lentiviral vector, and the luminescence value of Luciferase reaches 106 RLU after the pseudovirus infiltrates the cells, which can satisfy the neutralization detection and screening of antibodies or serums, and the results are accurate and reliable. In addition, Feifan can also undertake the packaging and neutralization testing of pseudoviruses for SARS, MERS and influenza viruses.
